How Peptides Actually Work: Signalling Versus Supplementing

The short version

  • A peptide is just a short chain of amino acids. Shorter than a protein, longer than a single amino acid.
  • Most therapeutic peptides are signals, not raw materials.

Signals, not supplements

The most useful mental model is that peptides are messages rather than ingredients. A protein shake supplies material. A peptide tells your body to do something it already knows how to do.

This has a practical consequence people find counterintuitive. If the machinery being signalled is impaired, the signal does not help much. Telling a pituitary to release growth hormone during deep sleep does very little if you are only getting five broken hours. The message arrives and there is nothing to act on it.

Why the ceiling exists

Signalling compounds work through existing feedback loops, which means those loops still apply. This is generally a safety advantage, since the body can decline a signal it does not need. It also means the effect size is smaller than supplying the hormone directly. Anyone marketing a secretagogue as equivalent to exogenous hormone is either confused or hoping you are.

Why the timelines are what they are

Why route and timing matter

Peptides are broken down by digestive enzymes, which is why most are injected rather than swallowed. Half lives are often measured in minutes, which is why timing relative to sleep and food is not a ritual but a pharmacokinetic requirement.

The one thing to take away

Peptides amplify a system that is already working. They do not replace one that is not. Fix sleep and training first, because they are free, and then decide whether you still need this.
