MOTS-c: What the Evidence Actually Shows

MOTS-c is not currently legal to compound. It was removed from the FDA restricted list in April 2026 and recommended by the advisory committee on 23 July 2026, but formal rulemaking has to complete first.

The short version

What it is

MOTS-c is genuinely unusual and that is part of why it attracts attention. It is encoded in the mitochondrial genome rather than the nuclear one, making it one of a small group of mitochondrial-derived peptides. It appears to act as a signalling molecule between mitochondria and the rest of the cell, influencing metabolic pathways including AMPK.

What the research shows

Animal work is the substance of it. In mice, MOTS-c administration has been reported to improve insulin sensitivity, reduce diet-induced obesity and improve exercise capacity in older animals. That is a genuinely interesting result set.

Human work is thinner. There is observational data associating circulating MOTS-c levels with metabolic health and exercise, but association is not the same as an intervention trial, and the intervention trials have not been run.
